Summary
We designed a cascaded spiral microfluidic platform. The microfluidic channel aspect ratio is different from the conventional fixed channel height design. This platform used different channel heights as the basis for adjusting the channel parameters. In order to optimized particle focusing conditions in the cascaded platform, we utilized the two-stage spiral micro-channel design with different heights, so that the small particles can be effectively removed in the first stage waste outlet without chemical added, while the large particles are in the second stage target outlet. Then, the system have high recovery efficiency to achieve the effect of pre-concentration of the sample. After previous verification, we apply the sorting of cancer cells in blood samples to pre-concentrate large number of cancer cells and reduce the interference of blood cells. Conclusion, this device provided a fast, high-throughput, low-cost and pre-concentration platform for cancer cell detection technologies.
Scientific Breakthrough
The unique designed of cascaded spiral microfluidic platform which separate principle only used the micro-channel structure to achieve the cancer cells enrich from the whole blood sample. The cancer cells can be pre-concentrated and a large number of blood cells are removed in different stage of platform. Experiment performed that our platform had high-throughput and fast to process samples. Besides, our device can reducing cell damage and improving detection accuracy without using high-cost drugs added, to achieve the purpose of significantly.
Industrial Applicability
The team's technology is a cancer cell sorting chip that can be used in clinically, with fast, high-throughput, and miniaturized characteristics. Compared with the existing technology, label-independent and no additional agent addition can provide a more complete cell type, beneficial to cancer analysis, diagnosis, and tracking. This rapid and label-free sorting cascaded platform is expected to be applied to precision medical technologies such as personalized medicine, providing fast and cheap cancer cell pre-concentration technology.
